Massive online scrabble site - when I looked there were around 5000 people online! And you can play in English (UK and US), French, Romanian, Italian or Dutch.
To get started you need to register (completely free) and then download Wordbiz (available for Windows, Mac and Linux), a program that allows you to play scrabble online. Once the program is installed you are prompted to open a connection (have your login and password handy) - its as simple as that! Click on "seek" to get a game.
If you want you can just observe a game. To get the full flavour of what you can do here click on the help tab.

Play Scrabble Online with others, Play Locally
To play at ScrabbleOn, you have to download the program Networdz.
To do this you need to join the games room - easy just click on the Scrabble On link on the left hand side, choose a user name and you are in. Type in the command !download and you will be given the download address. Type in the address into your browser and the download will start automatically. Install the program on your computer. You should then read the setup help section on the website in order to configure Networdz and to learn how to connect online.
Then you are free to play online against other players - you can also chat at the same time! There are tournaments and helpful tools such as word lists. It is completely free to play at ScrabbleOn. You can also play Networdz locally against your own computer.
You need to have Wndows in order to use Networdz.

Play Scrabble Online with others
What Yahoo has is a game called Literati - much like scrabble. It seems to be a very busy site - I had little trouble finding an oponent. Sign up (its completely free) and get started straight away. There are no downloads. The board is a little different to an "ordinary" scrabble board but operates in much the same way. There is a section on how to play or you can just hit the play now button (there is also a list of "rooms" further down the page that you can chose to play in and you can also choose between social, beginner, intermediate, advanced, ladder and league.

Single Player Game
Scrabble Blast is from Games.com. You can play it free online or you can download the paid for version. Although it has the feel of a Scrabble board, it is unlike the normal game in that you are presented with a board like the one shown on the left. You make words by clicking on letters, after which those letters are removed and the ones above drop down to take their place - with new ones filling in the spaces at the top. Scoring is similar to the usual game. Occasionally a letter tile will be a "bomb" and you have to use it in a word before it reaches the bottom of the board.
Beware! This game can be quite addictive ...

Play online against yourself. Very basic game but it plays nicely. The fact that there is no opponent means you can set up really good scores for yourself! This is a Javascript game so it shoould play in any Javascript enabled browser. It uses the UNIX dictionary.

There are no downloads and these are essentailly email games - you have your turn and your opponent receives an email telling them its their go a nice leisurely way to play! Braingamez has several word games and puzzle games to play. The one most like scrabble is Interwebword where you place letters on a grid - but in my view this is not as good as many of the other games listed on this page. But try it out - you might like it! It is another email word game with no downloads. You can find opponents on the site if you like or play with a friend.
